2aG4-IL-2 is an experimental immunocytokine consisting of the mouse monoclonal antibody 2aG4 fused to the cytokine interleukin-2 (IL-2). The 2aG4 antibody component specifically targets phosphatidylserine (PS), a phospholipid that is normally restricted to the inner leaflet of the plasma membrane but becomes exposed on the outer surface of tumor vascular endothelial cells and tumor cells in response to the oxidative stress and hypoxia characteristic of the tumor microenvironment. By binding to these exposed PS residues, 2aG4-IL-2 effectively concentrates IL-2 at the tumor site. This localized delivery is designed to activate and expand tumor-infiltrating lymphocytes, including CD8+ cytotoxic T cells and natural killer (NK) cells, thereby enhancing the anti-tumor immune response while potentially reducing the systemic toxicities associated with high-dose, non-targeted IL-2 therapy.
