2AP07 is a preclinical therapeutic vaccine candidate developed to target EpCAM-positive (EpCAM+) cancers, including head and neck cancer. The vaccine utilizes an adeno-associated virus-like particle (AAVLP) platform technology, into which mimotopes that mimic the EpCAM antigen are incorporated using phage display and next-generation sequencing selection methods. EpCAM is a transmembrane glycoprotein overexpressed in several epithelial cancers—such as breast, colon, head and neck, pancreas, and prostate—and is associated with cancer stem cells (CSCs) and circulating tumor cells (CTCs), both of which contribute to metastasis and resistance to standard therapies. By targeting EpCAM via immunization with the AAVLP-based vaccine displaying selected mimotopes at specific sites on the VLP scaffold, 2AP07 aims to elicit a robust immune response against EpCAM+ tumor cells while minimizing side effects. The project is being developed by 2A Pharma in collaboration with Danish Technological Institute (DTI) and DNASense[1][4][7].
