Overview

2b3-101 + trastuzumab is a combination therapy consisting of glutathione pegylated liposomal doxorubicin (2B3-101) and the monoclonal antibody trastuzumab. 2B3-101 is a brain-targeted formulation of doxorubicin encapsulated in glutathione pegylated liposomes, designed to enhance delivery across the blood-brain barrier and increase drug concentration in brain tumors. Trastuzumab is a monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2). This combination has been investigated primarily for HER2-positive breast cancer patients with brain metastases, aiming to leverage both cytotoxic chemotherapy and targeted anti-HER2 therapy. The mechanism involves DNA intercalation and topoisomerase II inhibition by doxorubicin (from 2B3-101), combined with HER2 receptor blockade by trastuzumab[1][4][8].
