2C11 x TA99 is a preclinical bispecific antibody (bsAb) designed to study T-cell redirection in syngeneic mouse models. It is constructed using Genmab's controlled Fab-arm exchange (cFAE) technology, combining a mouse CD3ε-targeting arm (derived from clone 145-2C11) with a mouse tyrosinase-related protein 1 (TRP1/gp75)-targeting arm (derived from clone TA99). The molecule utilizes an effector-silenced mouse IgG2a-LALA backbone to prevent non-specific Fc-mediated activation. By bridging T cells and TRP1-expressing melanoma or pancreatic cancer cells, it induces T-cell activation, cytokine release (IFNγ), and recruitment of innate immune cells like inflammatory macrophages and NK cells to the tumor microenvironment. This construct serves as a valuable tool for investigating the complex interplay between tumor cells and the immune system in a fully immune-competent host.
