Overview

The 2C5 antibody (mAb 2C5) is a research-stage, nonpathogenic antinuclear monoclonal antibody with nucleosome-restricted specificity, originally developed at Northeastern University. It specifically targets intact nucleosomes (chromatin fragments) that are exposed on the surface of various cancer cells as a result of neighboring apoptotic cell death, as well as within neutrophil extracellular traps (NETs). Rather than acting as a standalone therapeutic, mAb 2C5 is primarily utilized as a tumor-targeting ligand conjugated to the surface of drug-loaded nanocarriers, such as PEGylated liposomes, polymeric micelles, and dendrimers, to enhance their accumulation in tumors. It has been investigated preclinically for the treatment and imaging of multiple cancers (including brain, breast, lung, colon, and prostate cancers) and NET-associated conditions like heparin-induced thrombocytopenia and disseminated intravascular coagulation.
