Overview

`2E12` is an agonistic mouse monoclonal antibody (IgG1, κ) that specifically targets death receptor 4 (DR4, also known as tumor necrosis factor receptor superfamily member 10A or TRAIL-R1). Developed by researchers at the University of Alabama at Birmingham (UAB) in collaboration with Sankyo (now Daiichi Sankyo), 2E12 binds to cell-surface DR4 and, upon cross-linking, induces receptor trimerization and clustering. This triggers the recruitment of the death-inducing signaling complex (DISC), leading to the activation of caspases 8, 9, and 3, and ultimately inducing apoptotic cell death in various cancer cells. It has been investigated in preclinical research for its potential in treating malignancies and autoimmune diseases like rheumatoid arthritis, but has not entered clinical development.
