Overview

2G-1 TCR-transduced T-cells is an investigational autologous adoptive cell therapy developed by the National Cancer Institute (NCI) for the treatment of metastatic clear cell renal cell carcinoma (ccRCC). The therapy utilizes patient-derived T-cells genetically engineered via retroviral transduction to express a high-affinity T-cell receptor (TCR) designated 109Y-F/112S-K. This TCR is derived from the HC/2G-1 clone and specifically recognizes the TRAIL-DR4 (death receptor 4) complex, which is highly expressed on renal cell carcinoma cells. Unlike many TCR therapies that are MHC-restricted, this specific TCR demonstrates broad recognition of RCC cells across different MHC haplotypes, potentially allowing for a wider patient eligibility. The treatment protocol typically involves lymphodepleting chemotherapy followed by the infusion of the modified T-cells and high-dose aldesleukin (IL-2) to support cell expansion and persistence.
