2G10-PNU159682 is a preclinical **CDCP1-targeting antibody-drug conjugate** developed for RAS-mutant pancreatic cancer. It consists of the anti-CDCP1 monoclonal antibody 2G10 conjugated to PNU159682, a highly potent anthracycline-derived DNA topoisomerase II inhibitor. The antibody is intended to bind and internalize through CUB domain-containing protein 1 on tumor cells, enabling intracellular delivery of the cytotoxic payload and DNA-damage-mediated cell death. In published pancreatic cancer cell-line and xenograft studies, it showed antitumor activity in KRAS G12D- and KRAS G12C-mutant models.
