Overview

2G12-2B2-CL-MMAE is an experimental antibody-drug conjugate (ADC) designed to target tumor cells expressing the cancer-associated carbohydrate antigen **Sialyl-Tn (STn)**. It is composed of the monoclonal antibody 2G12-2B2, which specifically binds STn, chemically linked (via a cleavable linker) to the cytotoxic payload **monomethyl auristatin E (MMAE)**. Upon binding to STn on the surface of cancer cells, the ADC is internalized and releases MMAE intracellularly, disrupting microtubule formation and inducing apoptosis. Preclinical models show robust antitumor efficacy—especially in ovarian and colon adenocarcinoma— with high specificity for tumor tissue and minimal cross-reactivity with normal tissues. The agent demonstrates favorable pharmacokinetic and tolerability profiles in non-human primate studies[1][2][3][5][7][9].
