2G7 is a mouse monoclonal antibody (IgG2b) that specifically binds to and neutralizes high mobility group box 1 (HMGB1), a damage-associated molecular pattern (DAMP) and pro-inflammatory cytokine. It binds to an epitope within the amino acid region 53–63 of the A-box subunit of HMGB1, preventing its interaction with the TLR4/MD-2 receptor complex and subsequent pro-inflammatory cytokine release. Originally developed by Critical Therapeutics in collaboration with the Feinstein Institute for Medical Research and Karolinska Institutet, 2G7 has been widely used in preclinical research to study and treat HMGB1-mediated inflammatory conditions, including sepsis, rheumatoid arthritis, acute lung injury, and cancer progression (by reducing myeloid-derived suppressor cells).
