Overview

2L9V-GP2 (also known as SPA-12) is a modified HER2/neu-derived peptide vaccine designed to prevent breast cancer recurrence. It is a variant of the GP2 peptide (IISAVVGIL, residues 654-662 of the HER2/neu protein) with amino acid substitutions at position 2 (isoleucine to leucine) and position 9 (leucine to valine, specifically 2L9V) to enhance its binding affinity to HLA-A2 and HLA-A3 major histocompatibility complex (MHC) Class I molecules. By improving binding affinity, 2L9V-GP2 aims to elicit a more robust and functional cytotoxic CD8+ T cell response against HER2-expressing tumor cells compared to the wild-type GP2 peptide. The vaccine has been evaluated preclinically using peripheral blood mononuclear cells (PBMCs) from breast cancer patients.
