Overview

2MW4691 is an ADCC-enhanced bispecific antibody developed for cancer immunotherapy. It targets both C-C motif chemokine receptor 8 (CCR8) and cytotoxic T-lymphocyte-associated antigen 4 (CTLA-4). The drug is designed to selectively deplete tumor-infiltrating regulatory T cells (Tregs) by binding with high affinity to CCR8, a marker highly expressed on these cells within the tumor microenvironment, while exhibiting attenuated binding to CTLA-4. This dual targeting allows for specific elimination of intratumoral Tregs and blockade of CTLA-4-mediated immunosuppression on peripheral CD8+ T cells, thereby enhancing anti-tumor immune responses while minimizing systemic side effects commonly associated with broader CTLA-4 inhibition. Preclinical studies have demonstrated potent anti-tumor efficacy in humanized mouse models and favorable safety profiles in non-human primates[2][5][7].
