2MW4991 is a high-specificity, high-affinity ADCC-enhanced monoclonal antibody targeting integrin αvβ8. Integrin αvβ8 is an important activator of TGF-β, specifically regulating its activity in immune cells and being highly expressed in various tumors. By blocking the release of TGF-β mediated by αvβ8, 2MW4991 exhibits strong anti-tumor activity in multiple pharmacodynamic models (such as CT26 and EMT6), significantly promotes immune cell infiltration into tumors, and greatly increases the sensitivity of immune-excluded tumors to PD-1 inhibitors. The drug has shown good safety and metabolic profiles in non-primate models[6][3][4].
