**2NTX-99** is a novel small molecule anti-atherothrombotic agent developed by Nile Therapeutics for cardiovascular diseases. It inhibits thromboxane (TXA2) synthesis and action, enhances prostacyclin (PGI2) production, and donates nitric oxide (NO) to the vasculature, collectively inhibiting platelet aggregation, promoting vasodilation, and protecting against atherogenic stimuli. The compound showed preclinical efficacy in inhibiting arachidonate-induced platelet aggregation (IC50 ~10 µM) and reducing TXA2 production in rats after oral dosing, with potential utility in atherosclerotic, thrombotic, and microvascular conditions; however, its development was terminated in 2009 to focus on other pipeline assets.
