2R27 is a cross-species monoclonal antibody that specifically targets **CD74** (also known as the HLA-DR-associated invariant chain), a type II transmembrane protein highly expressed on the surface of antigen-presenting cells such as B cells, macrophages, and dendritic cells. Developed as a precision targeting tool, 2R27 is designed to bind to CD74 and undergo rapid internalization, making it an ideal candidate for the delivery of therapeutic payloads in **antibody-drug conjugates (ADCs)**. In the context of **systemic lupus erythematosus (SLE)**, 2R27 has been utilized as the targeting moiety for a novel conjugate known as **Bud-ADC** (or 2R27-Bud), which delivers the potent glucocorticoid **budesonide** directly to CD74-expressing immune cells. This targeted approach aims to enhance the therapeutic index of glucocorticoids by concentrating their immunosuppressive activity within pathogenic cells while minimizing the systemic off-target toxicities typically associated with conventional steroid therapy. Preclinical research in SLE mouse models has demonstrated that 2R27-based conjugates can significantly reduce autoantibody levels, alleviate splenomegaly, and mitigate kidney damage.
