3ε-EDB CAR is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the extra domain B (EDB) of fibronectin, an oncofetal antigen predominantly expressed in the tumor stroma and neovasculature of various solid tumors. Developed by researchers at China Pharmaceutical University and collaborating institutions, this construct is unique in its incorporation of the CD3ε subunit, which allows the CAR to displace native CD3ε and integrate into the endogenous T-cell receptor (TCR) complex. This integration enables the CAR to co-opt the natural TCR signaling machinery, potentially leading to more physiological T-cell activation and improved anti-tumor efficacy compared to standard second-generation CARs. Preclinical studies have demonstrated complete tumor regression in xenograft models of glioblastoma and significant activity against other EDB-positive cell lines.
