3ε28-EDB CAR is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the extra domain B (EDB) of fibronectin, an oncofetal antigen highly expressed in the tumor stroma and vasculature of various solid tumors. Developed by researchers at China Pharmaceutical University and Jiangsu Cell Tech Research Institute, this construct is unique in that it incorporates the CD3ε (epsilon) chain, allowing the CAR to integrate directly into the endogenous T-cell receptor (TCR) complex. Additionally, it features a CD28 intracellular domain to provide co-stimulatory signaling. In preclinical xenograft models, 3ε28-EDB CAR T cells demonstrated superior activation, proliferation, and tumor regression compared to standard second-generation CARs, effectively co-opting the TCR signaling machinery to enhance anti-tumor efficacy against EDB-positive malignancies.
