3-aminobenzoic acid (3-ABA) is a microbiome-derived metabolite and a structural isomer of aminobenzoic acid that functions as a gut barrier-protecting agent. It has been shown to decrease intestinal epithelial cell (IEC) permeability and increase trans-epithelial electrical resistance (TEER) in vitro. Research suggests that dysbiosis in patients with inflammatory bowel disease (IBD) leads to accelerated degradation of 3-ABA, resulting in significantly lower fecal levels compared to healthy individuals. In preclinical models of dextran sodium sulfate (DSS)-induced colitis, rectal administration of 3-ABA improved clinical symptoms, reduced histological damage, and suppressed the expression of pro-inflammatory cytokines. It is being explored as a potential therapeutic for restoring intestinal barrier function in IBD.
