Overview

3-chloro-5-hydroxybenzoic acid (3CL-HBA) is a synthetic small molecule that acts as a selective agonist for the hydroxycarboxylic acid receptor 1 (HCAR1), also known as GPR81. HCAR1 is a G-protein coupled receptor primarily known for being activated by L-lactate. In neurobiology research, 3CL-HBA is utilized to investigate the signaling pathways of lactate in the brain, particularly its role in modulating neuronal excitability in regions like the hippocampus and dentate gyrus. Studies presented at the AES Annual Meeting 2025 suggest that HCAR1 activation by 3CL-HBA can enhance baseline excitability and induce activity-dependent rebound responses in dentate granule cells, potentially influencing seizure susceptibility in conditions like temporal lobe epilepsy.
