Overview

3-glycosyl-3-odemethylthiocolchicine refers to a class of novel compounds that are derivatives of colchicine, specifically 3-demethylthiocolchicine with a glycosyl group attached at the 3-O position. These compounds can contain various natural and unnatural sugar moieties. They have been synthesized and investigated for their potential as ligands for gamma-aminobutyric acid (GABA) and strychnine-sensitive glycine receptors, found in the rat brain and spinal cord. The parent compound, colchicine, is a pseudoalkaloid known for its anti-inflammatory properties, primarily through tubulin disruption, inhibition of neutrophil chemotaxis, and modulation of NALP3 inflammasomes. Thiocolchicoside is a prodrug that metabolizes into 3-demethylthiocolchicine, which is then glucuronidated to form an active metabolite. The research into 3-glycosyl-3-odemethylthiocolchicine aims to explore new therapeutic avenues based on the colchicine scaffold.
