3-monothiopomalidomide is a structural analogue of pomalidomide in which a sulfur atom replaces one of the oxygen atoms in pomalidomide’s phthalimide ring system. It is not a standard, FDA-approved drug but rather appears in the scientific literature as a specifically modified derivative of pomalidomide, generally studied in the context of research on immunomodulatory imide drugs (IMiDs). Like pomalidomide, such derivatives are hypothesized to modulate the activity of the E3 ubiquitin ligase substrate receptor cereblon, thereby promoting targeted protein degradation and exhibiting potential antitumor and immunomodulatory effects. However, 3-monothiopomalidomide itself does not have published clinical trial data or regulatory approval. Its mechanism, extrapolated from its class, would likely involve binding cereblon to alter ubiquitin ligase substrate specificity, affecting downstream targets involved in cell cycle and immune regulation.
