Overview

CKBA (3-O-cyclohexanecarbonyl-11-keto-β-boswellic acid) is a novel small molecule derived from acetyl-11-keto-beta-boswellic acid (AKBA), a natural compound known for its anti-inflammatory properties. CKBA was chemically modified to improve efficacy and solubility for topical use, primarily targeting skin inflammatory disorders such as psoriasis. It acts by inhibiting Th17 cell differentiation through direct inhibition of acetyl-CoA carboxylase 1 (ACC1), thereby reducing the secretion and expression of the inflammatory cytokine interleukin-17A (IL-17A) in psoriatic lesions. Additionally, CKBA suppresses mast cell activation via the extracellular signal-regulated kinase signaling pathway and helps maintain skin barrier homeostasis. Preclinical studies have demonstrated its safety profile in animal models with no significant adverse effects observed upon systemic administration. The drug is being developed as a topical ointment for psoriasis treatment[1][2][3][4].
