3-TYP is a potent and selective small-molecule inhibitor of Sirtuin 3 (SIRT3), a mitochondrial NAD+-dependent deacetylase. By inhibiting SIRT3, 3-TYP disrupts mitochondrial metabolic homeostasis, leading to increased mitochondrial reactive oxygen species (mtROS) production and alterations in autophagy and mitophagy pathways. In preclinical models, particularly in triple-negative breast cancer (TNBC), 3-TYP has been shown to reduce spheroid growth and suppress the expression of oncogenic factors such as HIF-1α and c-Myc. It is frequently utilized as a pharmacological tool to investigate the role of SIRT3 in metabolic reprogramming, oxidative stress, and tumor progression.
