30F11-IgG1-C825 is an experimental bispecific fusion antibody designed for pretargeted radioimmunotherapy (PRIT) in murine models of myeloid leukemia. This construct consists of an anti-mouse CD45 monoclonal antibody (30F11) fused to the single-chain variable fragment (scFv) C825, which specifically binds to radiometal-labeled DOTA chelates such as ^90Y-DOTA-biotin. The primary function is to first direct the bispecific antibody to CD45-positive leukemia cells, followed by administration of a radiolabeled biotin molecule that is subsequently captured in vivo by the antibody’s C825 domain, facilitating targeted delivery of cytotoxic radiation to leukemia cells. This multi-step targeting is designed to improve therapeutic efficacy while limiting off-target toxicity. The approach has demonstrated increased survival in SJL murine leukemia models and pronounced uptake in bone marrow and spleen while minimizing exposure to normal tissues[1][4][5][7].
