313R12

313R12 is a surrogate anti-mouse TIGIT (T cell immunoreceptor with Ig and ITIM domains) monoclonal antibody developed by OncoMed Pharmaceuticals for preclinical research. It was generated to facilitate the study of TIGIT inhibition in murine models, as the clinical candidate OMP-313M32 (etigilimab) does not cross-react with rodent TIGIT. 313R12 blocks the binding of TIGIT to its ligand, poliovirus receptor (PVR), thereby inhibiting co-inhibitory signaling and enhancing the anti-tumor activity of CD8+ and CD4+ T cells and NK cells. In syngeneic mouse models, the antibody has demonstrated the ability to induce Th1 responses, increase cytotoxic T lymphocyte activity, and promote long-term anti-tumor immune memory, leading to complete tumor regression in colon and kidney cancer models.
