31B01 is a murine IgG1 monoclonal antibody targeting neuronal pentraxin 1 (NPTX1), a secreted protein identified as a critical driver of metastatic colonization in pancreatic ductal adenocarcinoma (PDAC). NPTX1 is significantly overexpressed in PDAC liver metastases and promotes tumor cell growth under hypoxic conditions by interacting with its cell surface receptor, AMIGO2. This interaction facilitates the nuclear retention and transcriptional activity of HIF1a. Developed through a collaboration involving Memorial Sloan Kettering Cancer Center and the Tri-Institutional Therapeutics Discovery Institute, 31B01 has demonstrated potent preclinical efficacy, significantly suppressing liver metastatic colonization and orthotopic patient-derived organoid (PDO) growth, while extending overall survival in mouse models.
