33BBzT2 is a preclinical chimeric antigen receptor (CAR) T-cell therapy developed by Wellington Zhaotai. It is engineered to target CD33, a transmembrane protein frequently expressed on the surface of myeloid cells and in hematologic malignancies such as Acute Myeloid Leukaemia (AML). The construct features a 4-1BB (CD137) co-stimulatory domain, which is designed to enhance the metabolic profile, persistence, and anti-tumor efficacy of the CAR T cells compared to first-generation or CD28-based constructs. The therapy is currently being investigated for its potential in treating Multiple Myeloma and Acute Myeloid Leukaemia.
