Overview

353T BCMA CAR-T is an experimental, monovalent chimeric antigen receptor (CAR) T-cell therapy designed to target the B-cell maturation antigen (BCMA), which is highly expressed on the surface of multiple myeloma cells. Developed by researchers at Fujian Medical University Union Hospital, the 353T construct was created as part of a comparative study to evaluate the impact of CAR valency on therapeutic efficacy, using the structure of ciltacabtagene autoleucel (Cilta-cel) as a reference. In preclinical evaluations, 353T demonstrated cytotoxic activity against multiple myeloma cell lines and in mouse tumor models, although it exhibited lower killing efficacy compared to bivalent constructs (such as 353/917T). Research indicates that 353T may induce stronger activation of the NF-κB pathway in target cells, potentially leading to increased expression of anti-apoptotic proteins like Bcl-2, which can be mitigated by combination with NF-κB inhibitors like SN50.
