Overview

3622W94 is a high-affinity humanized IgG1 monoclonal antibody targeting epithelial cell adhesion molecule (EpCAM, also known as 17-1A antigen or EGP40). Developed by Glaxo Wellcome (now GSK), the antibody was designed to treat various EpCAM-expressing epithelial cancers, including colorectal, gastric, lung, and prostate cancers, by inducing antibody-dependent cellular cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). However, clinical development was discontinued in Phase 1/2 trials due to severe on-target, off-tumor toxicities, specifically acute pancreatitis, which resulted from high-affinity binding to EpCAM expressed on normal pancreatic tissue.
