Drug intelligence / Profile preview
3A11 is a preclinical-stage, non-antibiotic antibacterial small molecule designed to combat multidrug-resistant (MDR) Gram-positive pathogens, including methicillin-resistant *Staphylococcus aureus* (MRSA) and vancomycin-resistant enterococci (VRE) [O0284, P2724]. It operates via a dual mechanism of action: primarily, it acts as an inhibitor of the copper-transporting P-type ATPase (CopA), which disrupts bacterial metal homeostasis and leads to toxic intracellular copper accumulation [O0284, P2724]; secondarily, it inhibits free thiol-containing proteins (such as glutathione) and induces protein aggregation [O0284]. Developed by a collaborative research consortium including the Karolinska Institutet, Université de Rennes, CIRI, Statens Serum Institut, and Maxel Consulting, 3A11 has demonstrated broad-spectrum bactericidal activity in vitro and is being evaluated in preclinical models.
