Overview

3A11-Exatecan is a preclinical-stage antibody-drug conjugate (ADC) targeting fibroblast growth factor receptor 4 (FGFR4). It is being developed by Manhattan BioSolutions in collaboration with the National Cancer Institute (NCI) and Binghamton University. The ADC consists of the high-affinity murine monoclonal antibody 3A11, which specifically binds to FGFR4, conjugated to an exatecan derivative (a topoisomerase I inhibitor) via a legumain-cleavable mpGlyAsnAsn linker. Upon binding to FGFR4 on the surface of cancer cells, the ADC is rapidly internalized and trafficked to lysosomes, where legumain cleaves the linker to release the exatecan payload, inducing DNA damage and subsequent cell death. 3A11-Exatecan is primarily being investigated for the treatment of FGFR4-expressing malignancies, including rhabdomyosarcoma (RMS) and breast cancer.
