3BNC117 + 10-1074 is a combination of two broadly neutralizing monoclonal antibodies (bNAbs) under investigation for the prevention and treatment of HIV-1 infection. Both antibodies target non-overlapping epitopes on the HIV-1 envelope glycoprotein, with 3BNC117 binding to the CD4 binding site and 10-1074 targeting the V3 glycan-dependent epitope. This dual targeting aims to enhance antiviral efficacy and reduce the risk of resistance compared to monotherapy. The combination has been shown in clinical trials to be safe, generally well tolerated, and effective at suppressing viral load during analytical treatment interruption in people living with HIV. It is being developed as a long-acting injectable for both pre-exposure prophylaxis (PrEP) and therapeutic use in individuals with HIV[5][6][7].
