Overview

**3BNC117-duoCAR-T cells** are an investigational, genetically engineered anti-HIV cellular immunotherapy developed by Albert Einstein College of Medicine and Caring Cross. The product consists of T cells modified with lentiviral vectors to express a multispecific duoCAR directed against conserved epitopes of HIV-1 envelope glycoprotein gp120 and to secrete the broadly neutralizing anti-HIV antibody 3BNC117, which recognizes the gp120 CD4-binding site. The duoCAR component is intended to identify and cytotoxically eliminate HIV-infected cells, while secreted 3BNC117 neutralizes HIV-1 virions and may augment control of viral spread. Preclinical studies in vitro and in humanized mouse models showed greater suppression of plasma HIV RNA and tissue-associated HIV DNA than either 3BNC117-secreting T cells or duoCAR-T cells alone. The program is being investigated as a potential strategy for durable antiretroviral therapy-free remission of HIV-1 infection.
