3BNC117-LS + 10-1074-LS + N-803 is an experimental combination therapy being investigated for the treatment and potential cure of HIV-1 infection. This regimen utilizes a 'shock and kill' strategy: N-803 (nogapendekin alfa inbakicept), an IL-15 superagonist complex, acts as a latency-reversing agent to 'shock' latent HIV out of infected cells while simultaneously activating natural killer (NK) cells and CD8+ T cells. The two long-acting broadly neutralizing antibodies (bnAbs), 3BNC117-LS and 10-1074-LS, then 'kill' the virus by neutralizing free virions and targeting infected cells for antibody-dependent cellular cytotoxicity (ADCC). 3BNC117-LS targets the CD4 binding site of the HIV-1 envelope glycoprotein gp120, while 10-1074-LS targets the V3 loop glycan-dependent epitope. The combination is currently being evaluated in Phase 1 clinical trials sponsored by Rockefeller University in collaboration with ImmunityBio.
