3C-rGel is a recombinant immunotoxin designed to target cells overexpressing fibroblast growth factor receptor 3 (FGFR3). It is composed of a human single-chain Fv antibody fragment, designated 3C, which provides high-affinity binding to FGFR3, fused to recombinant gelonin (rGel), a type I ribosome-inactivating protein (RIP). Upon binding to the extracellular domain of FGFR3, the construct is internalized via endocytosis and translocated to the cytosol. Once in the cytoplasm, the gelonin moiety enzymatically inactivates the 60S ribosomal subunit, irreversibly halting protein synthesis and inducing apoptotic cell death. Preclinical studies have demonstrated significant antitumor activity of 3C-rGel in xenograft models of bladder carcinoma, showing marked tumor growth delay and reduction in tumor size.
