Drug intelligence / Profile preview
3C-V1 is a novel proteolysis-targeting chimera (PROTAC) designed to target and degrade matrix metalloproteinase-7 (MMP-7). Developed by researchers at the Macau University of Science and Technology and Wuhan Union Hospital, 3C-V1 is being investigated as a potential therapeutic strategy for biliary atresia (BA). MMP-7 is a diagnostic biomarker and pathogenic mediator of biliary inflammation and liver fibrosis in BA. By degrading the intracellular MMP-7 pool in injured cholangiocytes, 3C-V1 limits subsequent extracellular secretion of MMP-7, reduces the secretion of inflammatory cytokines (such as IL-6, IL-8, and IL-17A), and reverses epithelial-mesenchymal transition (EMT)-related protein changes. In vivo studies in rhesus rotavirus (RRV)-induced BA mouse models have shown that 3C-V1 delays the onset of jaundice, reduces jaundice rates, and improves survival.
