3CAR is an investigational autologous chimeric antigen receptor (CAR) T-cell therapy being developed by St. Jude Children's Research Hospital. The therapy consists of a patient's own T cells that have been genetically engineered to express a CAR targeting the CD276 antigen, also known as B7-H3. B7-H3 is a transmembrane protein frequently overexpressed in a variety of pediatric and adult solid tumors while maintaining limited expression in normal healthy tissues, making it an ideal target for immunotherapy. Currently, 3CAR is being evaluated in a Phase 1 clinical trial (NCT04897321) for the treatment of pediatric and young adult patients with relapsed or refractory B7-H3-positive solid tumors. The treatment protocol involves an intravenous infusion of the engineered cells following a regimen of lymphodepleting chemotherapy to enhance T-cell engraftment and persistence.
