3D hypoxia-preconditioned umbilical cord-derived mesenchymal stem cells (UC-MSCs) are an advanced cell therapy product developed by the Chinese PLA General Hospital. Unlike traditional 2D-cultured MSCs, these cells are grown in a three-dimensional environment and subjected to low-oxygen (hypoxic) conditions during expansion. This specialized manufacturing process is intended to enhance the cells' therapeutic potency, survival, and secretion of immunomodulatory and hematopoietic growth factors by mimicking the natural physiological conditions of the bone marrow niche. The therapy is primarily being investigated as a sequential infusion in patients undergoing haploidentical hematopoietic stem cell transplantation (haplo-HSCT) for severe aplastic anemia, with the aim of improving donor cell engraftment rates and reducing the risk of graft-versus-host disease (GVHD).
