3D1015 is a next-generation radioligand drug conjugate (RDC) developed by 3D Medicines for the treatment of metastatic castration-resistant prostate cancer (mCRPC). It consists of a proprietary small-molecule PSMA inhibitor, 3D011, conjugated to the beta-emitting radionuclide lutetium-177 (¹⁷⁷Lu). The drug is designed to target prostate-specific membrane antigen (PSMA) with high affinity, delivering localized cytotoxic beta radiation to tumor cells to induce DNA damage and cell death. Preclinical data suggests that 3D1015 possesses superior binding affinity and tumor tissue exposure compared to the approved comparator Pluvicto (¹⁷⁷Lu-PSMA-617), while maintaining a favorable safety profile through rapid clearance from non-target tissues. It is currently undergoing Phase 1 clinical evaluation.
