Overview

3D6-k is a modified murine anti-amyloid monoclonal antibody, specifically a CDC-mutant (K322A) version of the 3D6 antibody. It is designed to block C1q binding, thereby reducing complement activation associated with amyloid-related imaging abnormalities (ARIA), an adverse vascular side effect observed in anti-amyloid immunotherapy for Alzheimer's disease. While maintaining similar fibrillar amyloid clearance capabilities as its parent antibody 3D6, 3D6-k aims to mitigate ARIA by reducing C1q/antibody colocalization in plaques and subsequent complement pathway activation.
