3D6-L is a monoclonal antibody that targets the N-terminal region (residues 1–5) of amyloid-β (Aβ) peptides, with neo-epitope specificity for a free aspartic acid residue at position 1. 3D6-L is structurally related to 3D6 and bapineuzumab (the humanized 3D6), exhibiting strong binding affinity for monomeric, oligomeric, and fibrillar forms of Aβ. It has been investigated primarily in the context of Alzheimer's disease for its capacity to reduce Aβ plaque load and related neuropathology. Preclinical studies confirm its ability to lower Aβ levels in animal models; however, administration in certain models has been associated with high rates of micro- and macrohemorrhages, indicating potential safety concerns, notably amyloid-related imaging abnormalities (ARIA)[1][2][3][4][6].
