Overview

3D9 is an agonist monoclonal antibody targeting the Thrombopoietin receptor (TPOR). Developed by researchers at The Scripps Research Institute and Zebra Biologics, it exhibits a unique property termed "receptor pleiotropism," where it induces a different cellular fate than the natural ligand, thrombopoietin. Specifically, in the context of acute myeloid leukemia (AML), 3D9 triggers the transdifferentiation of leukemic stem cells into functional natural killer (NK)-like cells. These induced killer cells express Granzyme B and Perforin, leading to the fratricidal destruction of neighboring AML cells. This effect is notably more potent in relapsed AML populations, which are enriched for stem-like cells. In healthy hematopoietic stem cells, 3D9 maintains the physiological function of TPOR by promoting megakaryocyte and platelet production without inducing the killer cell phenotype.
