Overview

**3E10-3G5** is a bispecific monoclonal antibody engineered to combine a cell-penetrating anti-DNA antibody (3E10) with an antibody (3G5) specific for MDM2, an E3 ubiquitin ligase. The 3E10 component enables the antibody to penetrate living cells via the equilibrative nucleoside transporter 2 (ENT2), delivering 3G5 intracellularly, where 3G5 binds to MDM2 and blocks its interaction with the tumor suppressor p53. This inhibition prevents MDM2-mediated ubiquitination and degradation of p53, leading to increased intracellular levels of active p53 and induction of tumor cell apoptosis. The antibody is under preclinical and early-phase clinical development strategies with primary indications including cancers (notably melanoma and ovarian cancer) characterized by MDM2 overexpression or altered p53 pathways[1][3][5].
