3E10-D31N (also known as GMABD31N) is a preclinical cell-penetrating monoclonal antibody fragment (scFv) developed by Gennao Bio using its proprietary GMAB technology platform. It is an engineered version of the 3E10 anti-DNA autoantibody, featuring a D31N mutation in the CDR1 region that enhances its nuclear penetration and DNA-binding affinity. The antibody utilizes the equilibrative nucleoside transporter 2 (ENT2) to gain entry into cells and localize to the nucleus. 3E10-D31N acts as a non-viral delivery vehicle by forming non-covalent complexes with various nucleic acid therapeutics, including mRNA, siRNA, DNA, and antisense oligonucleotides, facilitating their intracellular delivery. It is currently being investigated for applications in oncology and the treatment of rare monogenic skeletal muscle diseases.
