3E8 scFv is a humanized single-chain variable fragment (scFv) antibody that specifically targets Tumor-Associated Glycoprotein-72 (TAG-72), an oncofetal antigen overexpressed in various adenocarcinomas, including colorectal, ovarian, and breast cancers. Developed by Enlyton, LLC, this fragment is engineered from the humanized 3E8 monoclonal antibody to provide high tumor specificity with rapid systemic clearance via renal excretion, facilitated by its small molecular weight of approximately 28 kDa. It is primarily utilized as a targeting moiety for optical surgical navigation when conjugated to fluorescent dyes (such as IRDye800-maleimide) and as a delivery vehicle for nanoscale therapeutic platforms, such as polydopamine-stabilized nanotubes for the targeted delivery of camptothecin. Its high affinity and favorable pharmacokinetic profile make it a promising candidate for intraoperative tumor detection and targeted drug delivery in TAG-72-positive malignancies.
