This combination consists of two agents: - **3F8** is a murine IgG3 monoclonal antibody that specifically targets the ganglioside GD2. GD2 is highly expressed on neuroblastoma and some other tumors but has limited expression in normal tissues. The mechanism of action involves binding to GD2 on tumor cells and mediating antitumor effects through complement-dependent cytotoxicity (CDC) and antibody-dependent cell-mediated cytotoxicity (ADCC). Clinical studies have shown efficacy in treating high-risk neuroblastoma and minimal residual disease[2][6][9]. - **13-cis-retinoic acid** (also known as isotretinoin) is a synthetic retinoid derivative of vitamin A. It induces differentiation and apoptosis in neuroblastoma cells by activating nuclear retinoic acid receptors (RARs), leading to changes in gene expression that promote cell maturation or death[7]. The combination has been investigated primarily for consolidation therapy after initial treatment for high-risk neuroblastoma to eradicate minimal residual disease.
