Overview

3H1 (CeaVac) is a murine IgG1 monoclonal anti-idiotype antibody (Ab2) that serves as a molecular mimic of a specific epitope on the carcinoembryonic antigen (CEA). Developed as a cancer vaccine, 3H1 is designed to break immune tolerance to CEA, a self-antigen that is overexpressed in various adenocarcinomas, including colorectal and non-small cell lung cancers. By acting as an internal image of CEA, the antibody induces an anti-anti-idiotypic (Ab3) immune response that cross-reacts with the actual CEA on tumor cells, thereby stimulating both humoral and cellular (T-cell) immunity. The drug was primarily developed by researchers at the University of Kentucky and licensed to Titan Pharmaceuticals, but development was largely discontinued after failing to meet primary endpoints in Phase III clinical trials for colorectal cancer.
