Overview

3H2F CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy being developed for the treatment of glioblastoma (GBM). These cells are engineered with a T-cell receptor (TCR)-like antibody construct that specifically targets a TERT-derived antigen presented by the HLA-A*02:01 complex. The 3H2F construct is a modified version of the 3H2 clone, featuring a flipped orientation of the heavy and light chains (VH-VL vs VL-VH), which was found to preserve specificity while enabling potent cytotoxicity against GBM cells without the requirement for exogenous peptide. Developed at the Washington University School of Medicine, 3H2F CAR T cells have demonstrated effective in vitro tumor cell killing in preclinical models.
