3H3 is a rat-derived monoclonal antibody (IgG2a) that acts as an agonist for the 4-1BB (CD137) receptor. 4-1BB is a costimulatory molecule expressed on activated T cells and natural killer (NK) cells. By binding to 4-1BB, 3H3 provides potent costimulatory signals that enhance T-cell proliferation, survival, and effector functions, thereby promoting anti-tumor immune responses. In preclinical research, 3H3 is frequently used as a surrogate for human 4-1BB agonists to study the therapeutic potential of this pathway in murine models. Studies in multiple myeloma models have demonstrated that 3H3 can significantly reduce tumor burden and M-protein levels, particularly when combined with other immunomodulatory agents like tasquinimod, which helps overcome the immunosuppressive tumor microenvironment.
