3HP-2827 is a highly selective and potent small-molecule inhibitor of fibroblast growth factor receptor 2 (FGFR2), developed for the treatment of unresectable or metastatic solid tumors with FGFR2 alterations. The drug is designed to overcome resistance and reduce toxicities associated with less selective pan-FGFR inhibitors. Preclinical studies have demonstrated that 3HP-2827 selectively inhibits FGFR2 enzyme activity, showing over 1,600-fold selectivity compared to FGFR1 and FGFR3, and exhibits superior antitumor efficacy in models with FGFR2 alterations. It is administered orally and is currently being evaluated in Phase 1/2 clinical trials for safety, tolerability, pharmacokinetics, pharmacodynamics, and preliminary efficacy in patients with advanced solid tumors harboring FGFR2 mutations or fusions[4][5][6].
